Lantern Pharma (NASDAQ: LTRN), a clinical-stage, AI-driven precision oncology company, has secured regulatory and Institutional Review Board approvals in Taiwan to open Part 2 of its Phase 2 HARMONIC trial of LP-300. The lead Taiwan site is now open to accrual and actively screening patients, with additional IRB approvals anticipated at two more sites in the fourth quarter. In the United States, a central IRB has approved an amended protocol, and clinical sites have begun identifying patients with the EGFR exon 21 L858R mutation. This expansion is significant because it accelerates enrollment in a trial targeting a specific genetic subset of non-small cell lung cancer patients who often develop resistance to standard tyrosine kinase inhibitor therapies, potentially bringing a new treatment option to a population with limited effective alternatives.
Part 2 of the HARMONIC trial will enroll patients with the L858R mutation who have progressed after tyrosine kinase inhibitor treatment. The amended protocol allows LP-300 to be administered for up to eight cycles in combination with carboplatin and pemetrexed, an increase from the previous six cycles. Preliminary data showed a median progression-free survival of 8.9 months among nine L858R patients who completed six cycles, compared with 8.3 months among all 16 L858R patients evaluated. While the company cautions that these findings are preliminary, exploratory, and based on small patient numbers, the extension to eight cycles could further improve outcomes by providing longer treatment duration. This matters because L858R mutations account for a substantial portion of EGFR-mutated lung cancers, and patients whose disease progresses on TKIs have a poor prognosis with few targeted options. By leveraging its RADR artificial intelligence platform, Lantern aims to identify patients most likely to benefit, potentially streamlining development and reducing costs.
The Taiwan expansion also highlights the growing role of Asian sites in oncology trials, given the higher prevalence of EGFR mutations in Asian populations. Faster accrual could bring LP-300 closer to regulatory submission, offering a new therapeutic avenue. For investors, this progress validates Lantern's AI-driven approach and its pipeline, which includes LP-184, LP-284, and subsidiaries Starlight Therapeutics and Open-Medicine AI.
